spinoxin peptide neurotoxin consisting of 34 amino acid residues. has molecular weight of 3.7 kda. members of α-ktx subfamily share common cysteine-stabilized α/β motif (csαβ). in spinoxin n-terminal contains α-helix whereas c-terminal contains β-region/sheet, involved in binding potassium-channel, resulting in blocking of channel. 4 disulfide bridges connect these terminals stabilizing small protein. pattern of spinoxin disulfide bonding has been determined cys1-cys5, cys2-cys6, cys3-cys7, , cys4-cys8. 3d model of spinoxin can viewed on website.

spinoxin belongs class of small proteins knottin folds resulting 4 disulfide bonds. superfamily spinoxin part of known scorpion toxin-like , , family short-chain scorpion toxins .


homology

spinoxin has 82% sequence homology maurotoxin (mtx; α-ktx6.2). short-chain scorpion toxins contain 3 disulfide bridges, whereas several toxins belonging α-ktx6 subfamily, including spinoxin , maurotoxin, possess fourth disulfide bridge.
