pyruvate decarboxylation requires few cofactors in addition enzymes make complex. first thiamine pyrophosphate (tpp), used pyruvate dehydrogenase oxidize pyruvate , form hydroxyethyl-tpp intermediate. intermediate taken dihydrolipoyl transacetylase , reacted second lipoamide cofactor generate acetyl-dihydrolipoyl intermediate, releasing tpp in process. second intermediate can attacked nucleophilic sulfur attached coenzyme a, , dihydrolipoamide released. results in production of acetyl coa, end goal of pyruvate decarboxylation. dihydrolipoamide taken dihydrolipoyl dehydrogenase, , additional cofactors fad , nad+, regenerates original lipoamide (with nadh useful side product).
